Gene silencing: concepts, applications, and perspectives in woody plants Scientia Agricola

RNA interference, transcriptional gene silencing, virus induced gene silencing, and micro RNAs comprise a series of mechanisms capable of suppressing gene expression in plants. These mechanisms reveal similar biochemical pathways and appear to be related in several levels. The ability to manipulate gene silencing has produced transgenic plants able to switch off endogenous genes and invading nucleic acids. This powerful biotechnological tool has provided plant breeders and researchers with great opportunity to accelerate breeding programs and developmental studies in woody plants. This research work reports on gene silencing in woody plants, and discuss applications and future perspectives.

The aim of this paper was to compare the morphological attributes and production components of the wild-type Baronesa cultivar potato with those of Baronesa potato plants that have been genetically modified using the potato virus Y coat protein (CP) gene. The experiment was conducted under greenhouse conditions (Biosecurity Quality Certificate no. 081/98 issued by the National Biosecurity Technical Committee) at the Federal University of Pelotas (UFPel), Pelotas-RS. The shoot and root lengths and the mean leaf number were determined six times at 14-day intervals after sowing; the length, width and fresh weight of the tubers were measured during the last harvest, which occurred 84 days after sowing. European seabass is one of the most economic species in aquaculture in Mediterranean areas. Viral nervous necrosis (VNN), a disease affecting at least 70 aquatic species, has become the most serious threat to seabass cultured industry. While numerous studies have been performed in order to control this disease, no simple and effective procedures are available. Improvement of tomato for resistance to tomato yellow leaf curl virus (TYLCV) were carried out byusing a pedigree and backcrossing methods and crossing between resistant parent H-24 with susceptible parents CT-1 and CT-2. The virus was transmitted by viruliferous whiteflies and virus infection was detected by sandwich ELISA. F2 generation showed moderate resistance with scores ranging from 1.58-3.21. Segregation of disease resistance was observed among the population. Virus detection of the F2 populations (scoring 1 or 2) showed virus titer which were not statistically different from H24 but were significantly different from CT-1, CT-2 and susceptible control. Disease is a major problem in the swine production. The study of genes associated with disease resistance characteristics is used to select for innate immunity in swine. The Mx1 gene was found to have potent antiviral affect against some virus and can be hereditary. The study of Mx1 polymorphisms in among 3 pig breeds (Thai native pig breeds, Chinese pig breeds and European pig breeds) were found SNPs in exon 8 (c. 1022C>G; 1078A>C) that has allele frequencies of statistical significance between the breeds.
